I’ve been researching some of the new peptides in antiaging skincare products and am adding them to my Peptide Glossary.

These disorders include the aging process, obesity, metabolic syndrome, type II diabetes mellitus and Parkinson's disease.”In an article in the journal Nature, Harvard Medical School lead researcher Dr. David Sinclair reported, “It's looking like these sirtuins serve as guardians of the cell. For truly effective anti-aging treatment creams, look for peptides. You can do the research yourself and look at the before and after: 1. clinically-controlled photos; 2. silicone wrinkle imprints; 3. topographical maps of the imprints; and 4. mathematical calculations of decrease in wrinkle depth. Or you can let me tell you about them.
The science of peptide cosmeceuticals is in its infancy, so we can expect even more effective formulations in the near future. Finally, scientists have discovered how to deliver youth-giving ingredients deep into the dermis without injection.
Using penetration-enhanced peptides, creams can now replace ingredients which the body no longer produces in sufficient quantities or can stimulate processes which have slowed with the body’s decrease in hormone production.
There is a peptide proven to stimulate collagen production and plump wrinkles from within (Palmitoyl Pentapeptide-3). There is one which reverses glycation damage and facial sagging (Palmitoyl Tetrapeptide-3). Glycation damage stiffens collagen, stealing away the skin’s elasticity and the face’s youthful contours.
There is a peptide which interferes with transmission of nerve signals to frown and form deep facial creases (Acetyl Hexapeptide-3). The effect is cumulative. Nerve transmission signals are reduced about 30% immediately and after a month of twice daily application, wrinkle depth is reduced 40% on average.

Monday I quoted a gentleman who says he is an Associate Professor of Dermatology, Biochemistry and Biophysics, and Biomedical Engineering. He has been contributing to a thread on the Essential Day Spa forum. One statement he made raises his credibility in my mind.
“All anti-aging skincare is designed around anti-inflammation, as inflammation is considered to be the prime suspect in aging.” This is indeed a prominent theory of skin aging. Dr. Nicholas Perricone put it into layman’s terms in his best-selling books, The Wrinkle Cure and The Perricone Prescription. Dr. Perricone calls it the Inflammation-Disease-Aging Connection.
In our younger years, the inflammatory response is a good thing. It is the body’s way of walling off an injured area and preventing spread of infection. When its work is done, hormones shut off the inflammation.
However, as hormone levels decline with age, inflammation runs rampant. Older, hormone-depleted skin takes longer to heal and eventually doesn’t completely heal at all.
I find it very interesting that once you understand how skin care anti-aging ingredients function, you begin to realize that we are not just talking cosmetics and vanity, but that many such treatments can actually result in a better quality of life.
The list of skin anti-aging peptides is growing so rapidly that it is hard to keep track of them. To me, the most exciting yet are the tripeptides,
of which Synake is just one example. Whether Synake is “just” a topical anti-inflammatory or an ingredient that mimics the action of Botox, I am quite impressed by Pentapharm’s clinical study (see 6/14 blog entry).

While researching Palmitoyl Tripeptide-3, which I talked about Monday, I found research on a peptide that appears even more exciting at first glance.Palmitoyl Tripeptide-1 (PTri-1, Tradenames Trylagen and Aldenine) has been validated with in vitro, as well as in vivo studies using human subjects. The results are impressive. In vitro tests using reconstituted human skin, yielded increases in collagen I and collagen IV synthesis of 128 and 81% respectively after just 15 days. Collagen III synthesis (the most abundant type of collagen in young skin) tripled!Tissue sectioning of the treated skin tissue examined under an electron microscope revealed that the collagen fibers were significantly more uniform and better organized, as they would be in young skin. This effect results in fuller more supple skin.In addition, in vitro studies showed that PTri-1 even inhibits the body’s anticollagenase activity. Anticollagenase enzymes are one of the factors that cause collagen degradation as we age. Anticollagenase MMP-2 and MMP-3 were inhibited 74 and 57% respectively.In vivo, twice daily application of Trylagen by 20 female volunteers aged 35 to 55 for 30 days resulted in a 29% decrease in wrinkle depth as measured by 3D optical equipment.I’m on a quest for the best. I’ve just begun reading about Palmitoyl Tripeptide-3 and plan to research it more thoroughly. Cosmetic Design News had this to say just last month, “Tripeptide has become an increasingly important ingredient in recent years and [is] proving to be the ingredient of choice for anti-aging products, a fact that is highlighted by some of the most recent launches in the US.”Here are the basics, which I’ve added to my Cosmeceutical Peptide Glossary.
Palmitoyl Tripeptide-3 (PTri-3, Tradname SYN-Coll) is often referred to as synthetic collagen, because PTri-3 mimics the body's own mechanism to produce this youth-giving substance.
In vitro studies have shown PTri-3 to increase collagen synthesis by 119%. This makes it especially effective for parts of the face and body that have lost skin tissue, for example areas with stretch marks as well as areas with crepey skin such as often forms on the neck and upper arms. Its effect is to plump out the skin from within, similar to Palmitoyl Pentapeptide-3 (PP-3).
